PTSD poses a challenge in preclinical research and clinical practice. Interpersonal touch through mechanical stimulation of C Tactile afferents (CTs) can induce pleasant subjective experiences, activating brain areas that are involved in the processing of positive social-emotional feelings, along with behavioral, hormonal and autonomic responses. However, the neural mechanisms underlying PTSD as well as how CTs stimulation might contribute to PTSD treatment have not been fully elucidated. In this review, we focus on recent evidence derived from animal studies and human trials that implicate major brain pathways and structures, to help the reader understand how maladaptive cognitive, behavioral and sensory phenomena can manifest in PTSD, and how targeting mechanisms through CTs stimulation might improve quality of life of individuals with PTSD. This paper intends to instigate new discussions and research that could identify possible future directions for pre-clinical and clinical research in Manual Therapy for PTSD management.
