A anemia falciforme é a doença genética e hereditária de maior incidência no
mundo e, é causada por uma mutação pontual no gene que codifica a cadeia B-globina, isso faz com que seja produzida uma hemoglobina deformada e mais frágil, com tendência a sofrer hemólise. Portadores da anemia falciforme sofrem constantemente com crises vaso-oclusivas, infecções recorrentes, síndrome torácica aguda e acidente vascular cerebral. O único tratamento curativo é o transplante de células-tronco hematopoiéticas, mas o número reduzido de doadores irmãos, com antígeno leucocitário humano, compatíveis torna esse tratamento limitado. Recentes avanços na terapia gênica, com abordagens distintas, têm demonstrado grande potencial como cura definitiva para anemia falciforme.
Sickle cell anemia is the most common genetic and hereditary disease in the world and is caused by a point mutation in the gene that encodes the B-globin chain, which causes deformed and more fragile hemoglobin to be produced, with a tendency to undergo hemolysis. Patients with sickle cell anemia constantly suffer from vaso-occlusive crises, recurrent infections, acute chest syndrome and stroke. The only curative treatment is hematopoietic stem cell transplantation, but the small number of compatible sibling donors with human leukocyte antigen makes this treatment limited. Recent advances in gene therapy, with different approaches, have shown great potential as a definitive cure for sickle cell anemia.
